Principle and Procedure
- Precipitation: LDL, VLDL, and chylomicrons are precipitated from the serum by adding a precipitating reagent (e.g., phosphotungstic acid/magnesium chloride or Polyethylene Glycol).
- Centrifugation: The mixture is centrifuged to separate the precipitate, leaving HDL in the clear supernatant.
- CHOD-PAP Assay: The supernatant is treated with cholesterol esterase (CE) to hydrolyze cholesterol esters, and cholesterol oxidase (CHOD) to oxidize free cholesterol, producing
H2O2𝐻2𝑂2. - Color Development: H2O2𝐻2𝑂2 reacts with 4-aminoantipyrine and phenol in the presence of peroxidase (POD) to form a red quinoneimine dye.
- Measurement: The absorbance of the color, directly proportional to the HDL-cholesterol concentration, is measured at 505 nm.
